Biocoupler™ is the simplest temporary immersion bioreactor. It couples, connects, two mason jars (or any glass jars) with an integrated filter to facilitate a smooth medium flow. It also contains a microporous vent filter that allows pressure equalization to atmospheric pressure, thereby helping to prevent contamination. Moreover, the device is more efficient both space-wise and time-wise compared to other currently available TIB systems. Furthermore, the BioCoupler™ can be utilized not only by hobbyists but also by large-scale plant businesses.

- Biocoupler™ is used for temporary immersion and has been successfully tested for a wide range of plants, including orchids, hemp, aquatics, succulents, ferns, and carnivorous plants.
- It’s the simplest TIB system with a simple and elegant design, thus easy to operate.
- You don’t need to learn or have any prior experience before using Biocupler™ in your tissue culture system.
- It doesn’t require electricity to run.
- It’s more affordable and saves more space in your tissue culture lab. Thus, best for hobbyists, beginner culturists, or small-scale businesses. It’s also a great option for bigger labs based on their requirements.
- It works efficiently to multiply plants in hundreds or thousands under in vitro conditions.
- It’s a simple design and you only need to change its filter, which can commonly be found in any store, after some time—unlike traditional TIBs whose parts are often difficult to find.

How To Use
Biocoupler™
- Autoclave Biocouplers™ before use to avoid contamination. Utilizing PPM™ will help prevent contamination, benefiting plant growth.
- Select plant to multiply.
- Breakdown explants and prepare multiplication media.
- Add explants to sterilized Biocoupler™
- Rotate on a scheduled based upon plant species (typically between once daily to once weekly).
- Place Biocoupler™ in your growth area and monitor growth of explants.
- Transfer plants to the appropriate state (multiplication, rooting, or acclimation) based on lab's need.
The BioTilt™ (patent pending) automates the rotation of your Biocouplers™ at precisely timed intervals, optimizing the growth conditions for plant multiplication without manual intervention.
Plant Preservative Mixture (PPM™)
- General Dosage levels - With the exception of endogenous contamination, the recommended dose range is 0.05%-0.2%. (For callus proliferation, organogenesis and embryogenesis, the recommended range is 0.05-0.075%.) To eliminate higher endogenous contamination densities, higher doses of PPM™ are needed.
- (a) For explants: gently and routinely shake / stir 1 cm. long explants (or shorter) for 4-12 hours in 4-5% v/v PPM™ solution supplemented as above with 3x MS strength basal salts without Tween 20.Without rinsing, insert into a medium supplemented with 0.05 - 0.1% PPM™ for herbaceous plants and 0.2% PPM™ for woody plants. (b) F or ornamental plants only tubers, bulbs and scales: shake / stir the entire tuber/ bulb / scale in bleach. Rinse with water (can be done under non-sterile conditions). Slice the tuber / bulb /scale to thin slices. Shake / stir for 12-24 hours in 4- 5% PPM™ solution supplemented with full strength basal salts without pH ing & Tween 20. Without rinsing, insert into a medium supplemented with 0.1 -0.2% PPM™.
Murashige & Skoog (MS) Medium
Follow the given steps to prepare 1 liter of tissue culture media using MS media:
- Take a beaker and add 800 ml of distilled water to it.
- To the water add PCT’s MS Media 4.54 grams, 30 grams of sugar, and PPM™ (generally 1-2 ml/L)
- Make up the volume of the media to 1000ml.
- Adjust the pH of the media between 5.4-5.8.
- Add gelling agent (3-4 g/L gellan gum or 6-8 g/L agar) to the media and keep stirring.
- Autoclave the medium for 20 minutes at 15 psi and 121℃.
- Your media is ready!
Note: MS Media should be prepared according to the formula mentioned on the label. However, it is recommended to use an entire container at once.
